I think it would work out great. If your brother and his girlfriend get along well with you and his girlfriend's sister, then you could go on double dates and it'd be kind of perfect. But first, before making any move, you should talk to your brother and his girlfriend about it. You don't need their permission as such, but their advice might be helpful.
With the girl you like, if you're too scared to just make a move and see how she reacts, you could always bring up the subject as a joke. Ask her how she'd react if you asked her out, or if she thinks it'd be weird if you dated exclusively now that you know each other better. If her answer is negative, it's a bad idea to make a first move, obviously. If her answer is positive, you can either admit that you like her straight away, or wait for the right moment.

There's nothing wrong with doing that, and you only live once - so do it.
Just be yourself man, there is nothing to worry about =) You are all human and you dig her, so go for it.
You will not impact your brothers relationship; though if you want to be discreet - try to find your own ways of hanging out without being around them THAT OFTEN - it's okay to come around, though he probably wants his alone time too.
