If you want to lose weight as quick and safe as possible, then no, you should not eat it. Losing weight is one of those things that requires a substantial amount of sacrifice. There's always a special on donuts, or a buy 1 get 1 free pizza, or today only 12 patty death burger free one per customer. If you excuse yourself for stuff like that, then you'll always be overeating. I've lost 100 lbs myself and kept it off, and it's 80/20 diet/exercise. The diet part is essentially eating as little as possible without hurting yourself. An overweight person, according to me doctor, can easily survive on about 500 calories a day, though most diets recommend twice that. I have a very hard time eating only that much. What I do is have a snack as breakfast, another snack as lunch, and then a regular dinner, with coffee throughout the day. It never get's easy, but the first week is 100 times worse than the days after it. It's sort of like a diet hell week. If you can get through that, you essentially have it. But that means no excesses for that week. Cheat once, and it resets, because, life is unfair that way. Good luck!

Live life. Indulge in some sweets and junk every once in awhile.
The healthy and HAPPY people, don't live by excessively strict diets.
Eating fatty foods is the problem. You can't eat crap and then exercise and expect to lose weight. Changing your diet has a much bigger effect on your weight than any amount of exercise, so you would be setting yourself back by eating it.
You'll be fine. Assuming you modify your daily intake to account for the calories in that cupcake.

If you've been on a good regime the last couple of weeks just once shouldn't hurt, if you get right back in track. but if you're concerned you can always freeze it and eat it in a few weeks.
