Penis penetrating vagina obviously assumes a heterosexual couple. It's the most common definition, but obviously one that doesn't take full account of other orientations.
For gay and lesbian couples, I suppose it means "when someone else first brings you to an orgasm."
What are you "losing"? Mostly your sexual innocence and naivety, but for most women, it also means "losing" the intactness of her hymen, which for much of history was considered the "proof" of a woman's virginity. Keep in mind that into the 1960s, most women were married by 15-16, and sometimes as young as 12-13, so it was considered scandalous for her to not be a virgin on her wedding day, and non-virgins were generally seen as having much lower value as a result.
Things are MUCH different today, but the social mores are still based on those past standards, because we're only a couple of generations from those standards, and the people teaching the next generation are still trying to adjust.

some girls have a hymen (piece of skin around the opening of your vagina), and when a penis is inserted, or a dildo, it will break and bleed and hurt
having no hymen doesn't mean that you're not a virgin however, when girls are born, they sometimes break, doing sports, riding your bike, doing splits, can break your hymen

I do know what it means for most people but there are those that have a different definition like the above answerer. I asked the question to get an idea of what people think. I pride myself on being well informed, the first time I had sex, we used so much lube and I stretched out my hymen first so nothing tore, there was no blood and it didn't hurt too much. I think it's any sexual act for the first time. To me virginity isn't just a physical act, it's mental and emotional. So even just oral for me is losing your virginity it's about losing that innocence. There's too many factors if it's counted at penetration or breaking the hymen. Because then questions come up. Things like what if her hymen was not in tact? What if he doesn't break it because he doesn't enter all the way?
I would consider it to be the first time you are penatrated whether it be fingers, or penis by someone other than yourself. I would not consider self masturbation with fingers or toys loosing your virginity, but getting fingered by another guy or girl would count as loosing it. For a guy it would be first time he ejaculated with the help of someone else!

Of course not from your own fingers or for hygene or medical reasons, but I do consider it lost if it is someone such as a boyfriend or girlfriend who has their fingers in you!
