The main difference is, assuming her hymen is still intact, is that you have to "push through" a "seal" that is closing down her entrance to a smaller hole. That can be painful for her, as it usually tears, but some women have a minimal hymen or a very stretchable hymen and so sometimes it will go right in without much issue (the hymen may still tear a bit later on). At first, as you push through, you may feel the hymen making a tight ring around your cock, but that tends to go away because the hymen will usually stretch or tear at several points, but it might take 2-3 times before that feeling is completely gone.

The other thing is that virgins often struggle to relax, and so they can be difficult to penetrate for that reason, though in my experience, with a good amount of foreplay and getting her really turned on and wanting penetration, that's rarely a problem.
