If you dated someone for more than a year and they had a one night stand while on vacation out of state, would you stay or leave? Is it different since he/she would not have contact with the other person? What if you found that the one that you love has been spending a considerable amount of time with another, is that looked upon differently? I respect your thoughts here!

Get rid.
If its once or 50 times it was still a choice they had,they could of decided not to do it,and somewhere in the situation you (or whoever the person being cheated on is) would of popped into their mind...and it wasn't enough to stop them.
Different state? So what,could be a different country they still did it and I wouldn't care if they never saw that/heard from that person again its the principle.
Noway,cheating is cheating no matter how you try and wrap it up.

I would dump him. Once someone cheats on you, the trust goes out the window. If you take him back, you'll be worried every time he leaves the house/talks to women etc that he is in fact cheating on you. Once a cheater, always a cheater.

And how do you know he only a one night stand? Presumably because you're taking his word for it-not a good idea. You need to be more cynical and think "well well he would say it was just a one night stand to get himself out of this situation".

Both one night stands and long term affairs are both betrayal they're both equally bad. Frankly, if I found out someone had a one night stand, I would be thankful I had found out about earlier instead of later and just leg it out of the relationship. I would cut all contact with this person and just get on with my own life.
