Not really a spelling mistake, but "I could care less". It means the exact opposite of what people mean, ie "I could NOT care less". Unless maybe they are participating in a competition to see who could care the least.
Other than that, "then" versus "than", and also "they're", "their" and "there".
There are lots out there but a little think that gets to me is this site is in American English and a few of English words I use are spelt differently ( centre, center ) so when I key in my word the spellcheck screams at me.
"Your" vs. "you're".
Loose & lose. They mean two entirely different things, also, it's a grammmar mistake as they're two different words.
Lose - "I lost my phone."
Loose - "My pants feel loose."
