At first glance, without any extra info, I would say no that is not bullying unless it was repeated and the sister had said to stop. In that case, yes it would be. Bullying is constantly picking on someone or intimidating them. If that is what is happening, sure it could be considered bullying.

Now, I wouldn't take the comment about hand size seriously. I don't even know why that would hurt someone's feelings unless they were very self conscious about it. I have small hands. Never bothered me. If the sister is very aware of that and doesn't like to be teased, then that would be bullying by the younger brother. If that is the case, the sister should talk to her parents about it.
