One of my work colleagues said to me did you miss me while I was on holidays , and said I missed you while I was gone.
Is she doing this because she genuinely misses me and cares or just to mess with me

Most likely a shit test. An attempt to gauge to what degree she can manipulate you. Stay strong. You deserve more than she expects and she expects too much.
