Hmm. Probably Finnegans Wake. It's also one of my favorites, but any discussion of it begins with "it's one of the greatest books ever written, and you should probably never read it", and that just makes people want to punch you.

It is hard to find just one. I liked Dune by Frank Herbert and All Quiet on the Western Front by Erich Maria Remarque. Maybe the Sun Also Rises by Hemmingway or Travels with Charley by Steinbeck.
