A guy friend of mine recently asked if I was free to go out somewhere with him. This is the first time he’s asked to hang out 1 on 1 before in our 5 year friendship.

Am I right in thinking that this is a date and he may be interested in me?

Well you will only find out once you go out with him and see what setup it is... so I would just go and see what happens without putting any thought into it.

If it's really a 1 on 1 thing, it's probably a date. Unless it's you doing a favor for him, like helping him go furniture shopping or something.
