Most commonly, it would be interpreted as "give me some lovin'". That could mean a kiss, a make-out session, or even sex, depending on context and tone, but most commonly implies a sexy kiss and maybe a bit of making out.
Blues singer Nina Simone had a song in the late 60s called "I Want a Little Sugar in My Bowl" that was along the same lines.
Either someone needs some sugar on their food or someone's asking for some sweetness and love from someone else. It could mean some woman wants money from her sugar daddy or someone is referring to cocaine as sugar, a shortened version of booger sugar.
Usually it means give me a kiss but could be literal if a person is looking for some for their coffee or tea.
