He calls me sister but his behavior and how he treats me is confusing?

I have a close guy friend whom I hang out with and we talk a lot. He would say things like “i’ll always make time for you because i like you (as a friend)” and we hang out quite a bit. He’s very caring and takes care of me but recently he started saying “this sis” or “thank you sister.” I also use bro/dude a lot but i am not sure when a guy says sister, it means he is friendzoning you. What i’m confused is he would go down the length to make time to see me and the first guy that doesn’t say “i’m too busy” or ignores my request to hang out. He likes hanging with me so much that last year during Thanksgiving holiday he asked what i was doing and joined me and my family to our short road trip. by the way, this guy is super popular and has many friends so he has many people to have hanged out with during that holiday. And of course, he also asks me to hang out. Does my guy friend really see me as a sister?

I doubt that he actually sees you as his sister and instead uses terms like that to keep you in the friend zone. To me, this does nothing positive for you because if you like him more than just a friend, it will become hurtful having it frequently reiterated that you’re only friends. It will eventually sting when he’s talking about other women he’s taking interest in and wants your advice on. I feel like your current dynamic mainly works in his favor because he gets some girlfriend perks with you, like your time, attention and loyalty (to an extent), without deeper commitment by simply making sure you always remember your “friendly” place in his life. I don't know if that’s really all you want for yourself in the long run.

Guys do a lot of confusing stuff. The "sister" thing is probably a sort of "note to self" to indicate that you're a friend, which also tracks with his flip-flopping behavior. If he DOES like you... he doesn't actively WANT to (in that way). So just lighten up and don't push for more than what he's putting out there -- OR just distance yourself to leave your time available to those who respect and value you.
