Simply asking how someone is doing isn't a sign no matter if he never did it before or not and certainly not if it is a coworker. That is a normal exchange between people that work together and completely normal. If I took it to mean my coworkers asking me how I am doing as they like me everyone I work with has a crush on me then. They all ask that because it is a courtesy. Now about the fact that he hasn't done that before. Who cares? Maybe he was in a particularly chipper mood that day. Maybe he had some exciting plans for after work and was just happier than normal. Again... who knows but him?, The point is that saying how are you and reading that to mean he likes you is a huge leap.
