It depends on what sort of activities you like and the time of year.
The SoCal coast is amazing in the summer.
My wife and I spent two weeks in Massachusetts, New Hampshire and Vermont during the peak of fall one year. It was magical.
We recently spent a week in Albuquerque and Santa Fe, New Mexico, and had a great time in both cities as well as scenic sights and fascinating Indian sites. The balloon festival was also happening.
There's wine tasting and amazing wineries in Sonoma, Ca. There is also a cool place called Safari West there with free-roaming African animals (no predators) and safari ride tours. And there are redwood forests and great coastlines nearby.
If you're into theme parks, SoCal has Disneyland, Universal Studios, and Knott's Berry Farm.
There is Las Vegas.
And there are incredible and varied State and National parks all over the U. S.

I don't think there is one "best" vacation destination. If you want to immerse yourself in culture, New York City is probably the place to go. Want history? Then go to Boston, Philadelphia, or Charleston. Want wide sandy beaches with warm waters? The Gulf Coast beaches can't be beat.
Not so sure it's the best location but I really enjoy South Beach in Miami during the winter. The women are so much different down there compared to up north, I don't know if it's southern charm or wth.

I went to Nantucket a couple of timess. It was pretty nice.
