When I was on a dating site, it depended on what the message was from the person I wasn't interested in... typically decided from reading their profile info

If it showed that they had taken the time to read my profile, I would respond with a polite "Thank you very much but I don't think that we are a good match. Good luck in your searching."

If it was just a generic "Hi, how are 'ya" message, I would not respond at all.
