I don't think that she is trying to get at you for calling your mister.

I do also call males I am not familiar with "mister". I never call them by their first names. That would give the wrong impression. I am personally against calling people in general by their first names as it creates a closeness I don't want. Mainly men think that they can try something if a woman is calling them by their first names.

I like to keep my distance and calling a person Mr. XXX is my only way to go. I may switch to a first name once I know that person a little better but still keeping a healthy distance.

I don't know any details leading up to such a question but many girls & women address men with "Mister" as a form of respect.

Otherwise, if the girl you're referring said "Mister" with a sharp tone, she might have been in "defense mode" due to a discomfort she felt from you.
