I asked a girl I tutor a question and she answered it appropriately, but because her answer could be interpreted in a dirty way - another one of her colleagues said she had a dirty mind.. so she laughed, but when I asked her the same question again with a wink - she looked upset. Why was she upset? was I rude or was she rude and she can't take banter?

She might've only laughed out of embarrassment when the first colleague said she had a dirty mind, just trying to shake it off. And when you asked her a second time especially by winking, it may have made her feel like you weren't letting it go and trying to tease her about it, which could've made her feel more embarrassed.

I don't think sensitive, I just think she probably wanted you to drop it if she already felt embarrassed by the other person having to call her out like that by saying she had a dirty mind.

She didn't realize you knew the dirty part and was embarrassed. Trust me. I've had to explain to coed freshman what eunuchs, concubines, geisha girls, comfort women, hookers etymology, incest, fertility rites around the world, pederasty and God knows what else.
