It could mean one or both of the following:-
1) He has a soft voice in general, and is polite by nature.
2) He likes you.

Yes, it is possible but doesn't happen too often.

In my case, my voice is quite soft, and not completely masculine either. And I'm usually very polite with people unless they piss me off real bad. So there have been times when the combination of my soft voice and politeness have been mistaken for romantic interest by the other person.
