In normal conversations throughout the day you do things unconsciously to reduce tension - you avoid holding eye contact for too long, you smile, you laugh, you fill in the "awkward silences". Notice in the next few days how often you laugh at things that aren't even funny nor were they meant to be - you do this to reduce tension. Notice how quick you are to fill in those awkward silences. Why are they awkward? Because silence creates tension.
Tension can either be good or bad. For example, a man and another man (both straight) holding eye contact? That will be taken as a threat of aggression - bad. When it's a man and a woman however, and there's at least a little bit of attraction, and they lock eyes (which if they weren't attracted to you they wouldn't do, they'd look away) the tension is by definition sexual. And tension + attraction = arousal.

Sometimes, if the eye color is preferential and the male has already perceived a potential, attractive mate, it may be trigger a biological chemical reaction that sends blood down there if she is holding eye contact for more than 5 seconds.
