How long do we need to hold eye contact with you for you to know we like you?

This is probably one of my biggest issues when it comes to getting girls. I have an extremely difficult time maintaining eye contact with anybody, let alone an attractive girl. If it's a stranger, I tend to look away almost immediately, and if it's someone I'm in conversation with I can hold it maybe 5-6 seconds on and off.

My question is, in both scenarios (in conversation as well as just some guy checking you out), how long would a guy need to hold eye contact with you for you to have a pretty decent suspicion that he's into you?

Also just giving her multiple looks will give her a hint. Then if she's at all interested she'll start checking up on you if you're looking again! :) At least I do that haha.

I've managed to get that far on a couple occasions (multiple brief moments of eye contact, that is) but I never seem to get any smiles or acknowledgment beyond an unusual amount of peeks at each other.

Until I look down and look back up and see you still staring at me (but make sure to smile when she looks back up so you don't look like a creep)

she'd probably smile back, blush, or look away sheepishly(if she's interested) then you should say hi, keep it simple and sweet. And small talk should follow.

Any distinct signs that she's NOT interested?

she rolls her eyes (or anything rude of this nature), she flashes a quick smile (like a polite smile) and just continues doing what she's doing like if you don't exist.

I feel like a sheepish turn and a "not interested" turn would be indiscernible :/

if a girls looks nervous around you or shy it means she likes you. If she seems like you have no affect on her what so ever she is not interested.
