There are several, but, yeah, smoking, drugs, or more than occasional social drinking are deal breakers for me. But so are people who are selfish, shallow, rude, or unappreciative. Some people really think they are the main character of the movie, and will treat anyone they see as "beneath their level" like crap. I cut such people out of my life immediately.

And somebody who over talks a conversation especially when we're getting to know each other now both of y'all have the same personality naturally the conversation is going to go very smooth so my cut off is if all of her stuff is more important than mine has got to be an equal balance. Addictions, smoking (hate the smell of cigs), drug use (not heroin, meth, or crack) etc it depends on the individual and the circumstances, I don't use drugs personally but many in my circle do so I'm not perfectly clean.
