I’ve had it happen a few times in the time I’ve been dating my boyfriend but it is more on the lustful side. The one guy I flat out ignored. He was a coworker before I changed positions, knew I’m in a relationship, and sent me inappropriate messages after I switched jobs. The other was at a party and was an acquaintance. If someone I have no grudge against, but I'm not feeling it, that's rough. I'd be like: "Woah. Didn't see that coming. I may not be the best use of your time. Not feeling the same vibe. Best I let you know now, so you don't get hurt later."

If someone who's given me cause for a grudge, I'd be all Tobey Maguire like: "You should've thought of that earlier!"

If someone I have feelings back for, my thoughts would be: "Oh. Better not mess this up!" I'd try to play it cool, at first, until I gained more confidence.

Anytime someone has confessed any kind of feelings for me, those feelings weren't reciprocated. So, having to tell them that I don't like them in that way can kinda suck. Especially if I do like them as a friend because they're either going to bounce and I lose them as a friend or they might just stick around in hopes that I'll eventually develop feelings for them.
