This is an excellent question and there are quite a few I suppose.
I think they'd all involve Tom Hanks in someway with the possible exception of one...
1. From Cast Away...
I've got two scenes here that are sequential with a minor package delivery scene omitted here.
A) This is at the end of the movie. Tom Hanks' character, Chuck Noland, has returned from being stranded on a deserted island for 4 years. His fiancée, Kelly Frears, eventually moved on starting a family with another man. Still, he was the love of her life and his love for her is what kept him going. After reuniting at her home, she tells him that he was the love of her life and comes this close to abandoning her family to be with Chuck... but they both realize she has to stay with her family. So, Chuck lost her... AGAIN - first because of the plane crash and 4-year presumed-dead absence on the island and then right there when they realized they could not be together now.
Chuck then talks unloads to his buddy (whose wife died while Chuck was missing)...
B) After this scene, Chuck, a FedEx manager, hand delivers a package that was on the plane with him. Something that kept him going was to deliver that package, albeit 4 years late. The recipient is not home, but he leaves the package with a note. The package had an interesting butterfly image on it. His package delivered, he stops at a crossroads (below) in rural Texas wondering where to go next - both as a destination and with life. He coincidentally, but knowingly, meets the owner of the package who offers him directions. After she drives off, he realizes who she was and, at the very end, he has this wisp of a smile that ties in nicely with the scene above... "Who knows what the tide could bring?" (a reference to the tide bringing him a sail that enabled him to escape the island). He realizes that, even without Kelly, life has something to offer and is worth living.

A) The discussion between John Coffey and Paul Edgecomb (Tom Hanks) on the night before John's execution in "The Green Mile". John was wrongly convicted of murdering two little girls. He also has the ability to perform miracles and save lives. Paul is the head guard of this death row prison wing. Through a miracle touching of John to Paul, Paul knows that John is innocent and is a miracle worker. Paul is conflicted because he knows that John should be free and not executed. Paul genuinely fears God if he executes John and he says this:
" On the day of my judgment, when I stand before God and He asks me why did I... did I kill one of his true... miracles... what am I going to say? That it was my job... (*exasperated whisper*) it was my job? "
B) The ending. God HAS punished Paul by giving him very long life. I put two versions here. The first is for context, but the second ends better.

In both films, the title sequences set up the mood letting you know what you're in for... I am fortunate that I saw both these movies for the first time in theaters (1979, 1986) because movies like this (such as "Star Wars") NEED to be seen in theaters before seen on small screens at home.

This is the long establishing scene of the spacecraft Discovery en route to Jupiter. If you watch carefully and pay attention to details and think about how this was filmed, this alone deserved the 1969 Oscar for Best Special Visual Effects.

Wow, I've forgotten all about that film! Watched it like two decades ago, and I still remember much of the concept, so that means it left a mark. I'll make sure to watch it again soon.
As for mine, it's actually a tie:
1) Wanted (2008) - protagonist lashes out on a lethal roasting rant against his bitchy boss in front of entire crew, then packs up his personal keyboard and leaves. As he does, his "best friend" (who's actually fucking his girlfriend) lifts his hand up for a high five telling him "who's the man?". Protagonist then smashes keyboard into asshole's face. Camera goes to slow motion, screen shows broken keys spelling "fuck yo" with the final "u" spelled with asshole's broken tooth. Priceless. A soul purifier.
2) The Old Guard (2020) - two protagonists captured by military bad guys. Happen to be two men in love. As they comfort each other and kiss, one of the soldiers, typical highschool jock bully moron, makes fun of them asking "what, is he your boyfriend?" and all the soldiers laugh. One of the prisoners gives him such a demeaning scolding look (excellent acting) that it's felt through the screen, then goes on to belittle the soldiers for being so childish, bigot and pathetic. He explains what his partner, the love of his life, makes him feel, the sort of bond they have, and the soldiers' faces really show that they trully realize they will never in their lives experience such love. When he's done talking they actually lower their gazes and shift their weight in inconvenience because they're so ashamed, and the room is sileny. Game, set, match, absolutely beautiful writing, acting and directing. Movie itself ain't half bad, another Matrix trying-to-be, but this scene makes it superb.
