Whenever... There's no rules. I'm sure there's many shows and movies where a trans character has a lead role. Noteablely Hunter Schafer as Jules in Euphoria, Yasmin Finney as Elle in Heartstopper, Laverne Cox as Sophia in orange is the new black. They all had somewhat lead roles, episodes where it was mostly based on their story.
