Well Matlock used to be Sheriff of Mayberry is a past life and Mason is in a wheelchair, so I'd give the edge to Matlock.

Mason is the more intellectual and analytical attorney, while Matlock is folksy and intuitive. What kind of case is it and what is the venue? Big city or small town? Jury trial or trial to the judge?

I've heard that Andy Griffith was an amateur boxer, so Ben would win a fair fisticuffs fight.
But I don't think that's what you mean.
In court, they each had an excellent support staff that, somehow, always revealed the one piece of evidence that was needed to win the case.
Perry was a more polished traditional lawyer while Ben was more of the friendly let's investigate type, which would probably do better in today's courtrooms.

Perry Mason. I saw a documentary about Andy Griffith, he suffered from some paralysis from Guillain Barre syndrome. It's a horrible illness.

In real life these guys were the opposite of the characters they played on tv. Andy Griffith was a real mean bastard and Raymond Burr was a big old pillow biter.
