Is saying I alreasy have someone less painful than I'm not interested?
I think it would hurt either way but what would be the best thing to say or do in such situation?

Only if it's the truth. Just think of it in the reverse, what would you want to hear. You don't want a lie do you? You can't avoid hurt or disappointment, but you can make it less by not making him feel bad for asking. Be nice, but also give him the respect of being honest with him. Just simply say that you're really glad he asked, but you're very sorry you're not interested.
You don't want them to say they're not available, and then find out that they lied to you. Even if it was to "protect" your feelings, it's disrespectful right? You'd feel that way, so will he.
She has no obligation to tell the truth. He is not entitled to any information about her personal life or her reasons for not wanting to date him. "Thanks, but no." should be enough but too often it is not. For their own safety, women tell men whatever will make them go away with the least fuss. "I'm seeing someone" works very well, because the man can't argue with that.
"I'm seeing someone" is like "I'm washing my hair that night" and many other social fictions. People with a clue understand that they aren't necessarily true, but provide a way to reject someone without giving a specific reason. Such fictions are far better than the truth which would often bey something like "Sorry, you seem rather stupid", "No, you are too short/fat/ugly/nerdy/whiny/awkward/boring/annoying/unambitious..."
I like you so I don’t want to mistreat you and leading you to believe that I have same feelings as you…I would not do that to anyone I like. 20-30sec of of soak-in awkward time.

It's impossible to prevent disappointment. Don't try. Tell them whatever will make them go away with minimal fuss. "I'm seeing someone" works well, because it has nothing to do with their desirability.
