Acquaintance has a hot friend. How do I ask through her if he’s single? How can I ask her to set me up with him?

First, how well does this acquaintance know this guy?
You say this person has a 'friend' so if they know him well, you may have to get to know this acquaintance more. there's a decent chance that through casual conversation you can learn some more about this guy. Shoot, you may even be able to find him on some social media with a little effort and find out right now.

Second, can't you try making 1 on 1 conversations with the guy either in person or through some messages yourself?
If you have anxiety or something, it's cool, just try to be friends with this acquaintance and you may end up in a circle of friends which usually helps in things like this.

Maybe ask her if you could get to know him a little bit or something or just ask “can you try to set me up with him”
