No of course not. Why is someone offering to do a favor considered rude? He/she is just trying to be nice. Might be their way of connecting with employees. Whatever the case is they are not being rude. It is quite the opposite of that.

It would be rude, if you said - yes please, and then never drink that coffee or even worse - pour that coffee into a toilet ant he accidentally saw it. So, no, its not rude to say no.
