It depends on if you're trying to go for a good time or for fun. If you're going to go for time, you should do sprints to increase speed; run for 5 or 6 miles to build some stamina. Then, run a 5k to see where you are. Drink plenty of fluids. Pilates may help you look good but it doesn't help with running; running is a whole different thing. It's similar to swimming. The only way to improve is to do it.

Start with slow runs of 2 or 3k, then do intervals (fast laps, then slow laps, then another fast lap) for 5k. As you get stronger, do more fast laps than slow ones.
It doesn't take long to get ready for a 5k if you are used to working out. You won't set any records, but you can come through it without having to walk any part of the 5k.

How much time do you have until this run?
I'd start out with a mile then increase it by a 1/4 or 1/2 weekly depending on your time frame.
If you can do more then a mile without killing yourself then do so & build up from there. There are some apps for smartphones/ipods that do interval training to get you ready for a 5k (Couch 2 5k). Try those. They are great.
