It's a form of flirting. So, if you're having a conversation and you're both enjoying it, but all of a sudden, she goes: "How are you single?" then that's flirting... it's more of a rhetorical question in that aspect. But if she is asking you why you are single, then she's expecting you to answer it.

You could just say that you're single by choice or why you chose to be single. You don't have to delve deeper into it, but I think she is just trying to get to know you. If you're not over your ex or something like that, she might not pursue anything with you.

Say you were busy or you were taking care of family or you were working on your goals or you relocated and had to separate from your previous girlfriend or you had health problems and were avoiding women or you did not have time for a social life so never met enough girls to build a relationship or you liked the same girl for 8 years and never shared that time with other females.
