I haven't been on any dates nor had a girlfriend, there have been flings/one night stands here and there.

If I try to build something from a friendly route with a girl making her laugh, and cheering her up she'll eventually say she only sees me as a friend.

Make a list of your strong/weak points and start by finding remedies for your weak points while you enhance your strong ones. Caution though. What you perceive as strong points may be seen as negative ones by somebody else as like in everything revolving around humans, it is all relative.

To answer your last question first, both of the approaches you mentioned can work, so you might not be doing anything wrong but just haven't found the right person yet. I don't think you can make chemistry happen. You can increase it or reduce it by your actions, but it's either there or it's not. There's no secret formula to having chemistry with someone. You just have to keep trying until you find someone you have it with. Sometimes that takes a frustratingly long time.

To answer the original question, I think you know you have it when the two of you just feel totally at ease with each other and you both are excited to be with each other and can talk for hours about all kinds of things and both feel sexual attraction to each other.
