I haven't been on any dates nor had a girlfriend, there have been flings/one night stands here and there.
If I try to build something from a friendly route with a girl making her laugh, and cheering her up she'll eventually say she only sees me as a friend.
If I start off from a flirtatious route and make her laugh, shed be down to have sex but no matter how much I can impress her/compliment her/ offer* take her out its not enough to build a relationship..
Tons of attempts and no one has considered actually trying a relationsuip with me and my character isn't mean or needy at all, I've been told im an 8/10 at most.

Chemistry is something you need to build up. Being natural and not somebody that is not you is not going to work since you cannot keep that state for ever.
Make a list of your strong/weak points and start by finding remedies for your weak points while you enhance your strong ones. Caution though. What you perceive as strong points may be seen as negative ones by somebody else as like in everything revolving around humans, it is all relative.
If you are insecure, rehearse your appearance in front of a mirror. Perhaps that a change in the way you present yourself, a new hairstyle or new wardrobe can help you out there.
Important is not to be afraid to fail because you will fail initially.

To answer your last question first, both of the approaches you mentioned can work, so you might not be doing anything wrong but just haven't found the right person yet. I don't think you can make chemistry happen. You can increase it or reduce it by your actions, but it's either there or it's not. There's no secret formula to having chemistry with someone. You just have to keep trying until you find someone you have it with. Sometimes that takes a frustratingly long time.
To answer the original question, I think you know you have it when the two of you just feel totally at ease with each other and you both are excited to be with each other and can talk for hours about all kinds of things and both feel sexual attraction to each other.
It's a little hard to describe really, but it's a wonderful feeling and I think you'll know it when you have it.
I've tried that but theyre more down to have sex than be with me. there's a saying that girls aren't looking for guy friends. Most of interests involve computers and playing instruments but im not well versed in certain artists and music Im very open so I don't know how to relate 🤔

Chemistry really comes naturally. You seem to be trying to make someone like you. That's not building chemistry, but trying to make them feel comfortable around you. Chemistry is a feeling which makes both you and the other person feel comfortable around each other, without any effort.
