There is this coworker I like who generally flirts a lot with me in person but we never exchanged numbers since we are in different departments and could never find a reason to ask each other for our numbers. He was sick and so I took his number from one of our colleagues to send him a get well soon text. He responded nicely and saved my number as well. Have I come across as desperate? What do you guys generally think of girls who make the first move?

If the guy likes the girl then it is a good thing because it makes him feel more confident that she feels the same way. And don't worry, you haven't come off as desperate. If you were to start flirting with him in real life or ask him to go for a coffee, that would come off as desperate behavior. Making the first move on text was a good decision.
