I'm seeing a lot of people doing this now. I wouldn't think that was the best way to lose weight, possibly quicker.

With the added weight it's not even important to run, walking will be pretty effective under the circumstances.
But the weight should help for the weight loss aspect because weights are more benificial to losing weight cause it goes after the fat as its energy source, whereas cardio activities like running actually use muscle as an energy source and that's why people don't get bigger from running like they do weight lifting.

I wouldn't do a lot of running with a biacpack since your knees and back are vulnerable when you change the weight distribution. They might start hurting-in fact, almost certainly WILL start hurting...if you do that very much.
Long cardio sessions, long runs, will do you fine for weight loss. If you want a stronger back, do weights!
Well your body burns more calories to support the extra weight so it would help make your workout more effective. Of course if all the extra weight is in a backpack that can really screw up your back. It would be better with a weight belt or something like that.
You're going to put so much stress on your back doing that. You'll burn more calories but I highly suggest you do both separately. Do weight training before or after your run. Back problems aren't something you want.

It would be much easier and safer to go to the gym. Walking around with a weighted backpack all day everyday puts way to much stress on your back and legs. Just develop a full body workout scheduele that you will follow every week at the gym, that's the best way.
The more you work out, the more you lose. Of course running with a weighted backpack will help you lose weight faster. Keep in mind that you run the risk of injuring your back or your legs doing things like this. "How much more...?" would be impossible to answer because weight loss is due to your metabolic rate. The only answer is "more".
I heard of a guy who ran with a back pack and for every lb he lost he put one lb in the backpack tell the backpack had X amount of weight he wanted to loose. I think it's a good idea, I hate running/walking so if I wanted to loose weight by walking I would defiantly do the backpack thing.
I walk everyday and twice a week I use a backpack. The walk with the pack is more strenuous and I'm sure I am burning more calories. I am not so sure about running with a full apck. Sounds like you could hurt yourself.
I think its trying to emulate exercises such as weighted squats and lunges.
However, all that weight puts stress on your back. One is better served doing resistance training on an elliptical.
