They took off the profile picture of them together and his wife just put a selfie of herself and he put a car up should that be concerning or it don’t mean anything?

It's kinda weird, but there could be a legit explanation. If a professional photographer took the wedding pictures, the photographer owns the copyrights to the pictures. It's possible the photographer saw the picture (s) posted, which is a violation of copyright, and asked your brother and wife to take them down.
