Male friend (and a colleague) asked me whom I'm on vacation with. Does it have a meaning?

Yep... he called me, I declined and told him I'm traveling and will call him later. He said "ok".

Then I messaged him hours later saying "I'm really tired today... cannot talk". He asked "whom are you with there?"... asked twice. I said "family". And he replied "who exactly?"

Anyway... I wonder why is he so curious about it?

That's a little invasive of him to be asking who you're with, and you didn't have to answer that either. Asking "who exactly?" is very pushy and not his business. But if he was asking that much it's probably because he wanted to know if you were with a guy, because he might like you.
