Depends if you just want to get the challenge done, or make your legs&ass better. To work them out properly so that they grow, it's best if you wear them out, to the point you're barely able to walk afterwards lol. For workouts sake best is 3-5 sets of squats, each set until the point your legs surrender. Rest time in-between sets ca. 2minutes.
If you give your body enough time to recover, like half an hour or more in-between, the workout becomes by far less effective. So your somewhat dumb fitness trainer brother was right.

I don't know but the workout is much harder if you do it all at the same time so if anything there is truth to what he says. think about it, what if you did like 1 squat every 20 minutes? wouldn't be much of a workout, although that example is drastic hahaa.
I accidentally hit false but your brother is right it's better to do them all at once than at different times.

It would be advised to do other exercises along with squats, as squats are a compound exercises they work various parts of the body but do not focus on any one part to much. I would only do the squats as long as you can keep strict form and go slow as the more you tire yourself the more wobbly, unbalanced and weak you become if you can't do them right then you will only end up hurting yourself. Go only to your limits don't push yourself to hard.

do as much as you can do at the same time, but do not force additional squats upon yourself, you will kill your back and overacidificate your muscles - which will harm your success.
generally don't only do squats, push yourself, but not to hard and - most important - do the exercises right! you will not only not succeed if you don't, you will harm yourself, too.Just do weight training instead.
